- Create a new dataset (new or edc). Choose pulse program zg30 to record a conventional 1H spectrum.
- Remember to choose your own folder. Select ZGESGP on the Experiment (Same as mentioned before – ZGPR or PROTON).
- Type getprosol
- Check o1p: Set the offset to 4.7 ppm (H2O + D2O)
- Calibrate the pulse (pulsecal)
MODIFY SPECIFIC PARAMETERS
- Increase/Decrease the sw as required.
- You can adjust the water suppression using gs and adjusting SPdB1 (see picture below).
- Calculate the gain by typing rga – after the optimization of the water suppression.
- To start acquisition: zg
- The expected experimental time is displayed with the expt command).
PROCESSING
The recorded data is Fourier transformed with ft (or ef) and phase and baseline corrections are performed using apk and abs, respectively. For proteins, better use qsin combined with ssb=2.
Use .ph to adjust the phase.
